Blocking Neuropeptide Y1 Receptor May Ease Osteoarthritis by Restoring Cartilage Cell Cleaning Through a Key Cell Survival Pathway

Abstract

NPY and Y1R expression were significantly elevated in human osteoarthritis (OA) cartilage compared to normal tissue.

  • NPY treatment at 5 μM suppressed chondrocyte proliferation and increased MMP13 production while reducing COLII expression.
  • Activation of the PI3K/AKT/mTOR pathway was observed following NPY treatment, accompanied by inhibition of autophagy-related genes.
  • In vivo NPY injections induced OA-like changes, including cartilage thinning and mechanical allodynia.
  • Treatment with a Y1R antagonist reversed NPY's effects, restoring autophagy and reducing cartilage degradation.
  • Y1R antagonist treatment also improved gait parameters and reduced subchondral bone sclerosis in the OA model.
