Urolithin A may protect brain cells in Alzheimer's by reducing harmful cell recycling through VDAC1

Abstract

Urolithin A (UA) improved cognitive dysfunction and reduced Aβ deposition in APP/PS1 mice.

  • UA activated autophagy and increased levels of autophagy-related proteins in both APP/PS1 mice and Aβ-injured cells.
  • Phosphorylation levels of the PI3K/AKT/mTOR pathway were down-regulated, while AMPK phosphorylation levels were up-regulated in the presence of UA.
  • UA down-regulated VDAC1, which is associated with its neuroprotective effects against Alzheimer's disease.
  • The activation of autophagy and modulation of the PI3K and AMPK pathways by UA could be reversed by VDAC1 overexpression.
