Abstract

Alzheimer's disease (AD) is the leading cause of dementia worldwide.

  • AD is characterized by progressive synaptic dysfunction, neuronal loss, and cognitive impairment.
  • Current treatments only provide symptomatic relief and do not alter disease progression.
  • Neurosteroids, such as allopregnanolone, may have therapeutic potential by regulating neuronal function and neurogenesis.
  • These neurosteroids are associated with neuroprotective effects that may reduce amyloid-β accumulation and tau hyperphosphorylation.
  • They could enhance neuroprotective autophagy and activate signaling pathways related to AD, such as AMPK and BDNF.
  • The precise role of neurosteroids in the context of the mTOR/AMPK/BDNF signaling axis in AD remains unclear.
