Self-Amplifying mRNA-based Virus-like Vesicles In vivo Generation (SAMVIG) enhances antitumor efficacy in multiple tumor models.

  • SAMVIG facilitates the in vivo production of virus-like vesicles using a single mRNA construct.
  • The system activates the immune response through dual antigen presentation and immune modulator co-expression.
  • Local and systemic administration of SAMVIG led to increased recruitment and activation of tumor-infiltrating lymphocytes.
  • Robust antitumor efficacy was observed in both subcutaneous and metastatic tumor models.
  • SAMVIG allows integration of checkpoint inhibitors and other proteins to improve therapeutic outcomes.
  • The system demonstrated a favorable safety profile in vivo.
