Using Nile Red Fluorescence to Measure LC3 Protein Binding to Cell Membranes

Updated

Abstract

Essence

fluorescence provided a simpler way to monitor LC3 protein binding to lipid bilayers.

Evidence

An in vitro protein-lipid binding assay showed LC3 caused concentration-dependent decreases that correlated with ultracentrifugation measurements, with cardiolipin and ceramide facilitating binding.

Caveat

The method cannot provide absolute binding values and was tested as a biochemical bilayer assay rather than a cellular autophagy readout.
