Nilotinib reduces inflammation and cell death in brain immune cells by controlling NF-κB signaling and boosting cell cleanup and membrane repair

Abstract

Nilotinib significantly attenuated NLRP3 inflammasome activation in murine microglial cells.

  • Reduced NLRP3 expression and decreased caspase-1 activation were observed with nilotinib treatment.
  • Secretion of proinflammatory cytokines IL-1β and IL-18 was suppressed through modulation of the IκBα/NF-κB signaling pathway.
  • Nilotinib inhibited pyroptotic cell death by decreasing GSDMD-N and NINJ1 expression.
  • Enhanced BAG3-dependent selective autophagy and increased LC3B expression suggest activation of pathways for clearing inflammasome-related components.
  • Restoration of VPS4A expression indicates activation of mechanisms for plasma membrane repair.
