New niosome method for effective gene delivery in aging umbilical cord stem cells

Updated

Abstract

DP20CQ niosomes prepared at a 10/1 DOTMA/DNA ratio achieved the highest transgene expression levels in senescent umbilical cord mesenchymal stem cells.

  • Niosome-based gene therapy shows potential in overcoming limitations of first-generation senotherapeutics.
  • Two new niosome formulations were developed to genetically modify senescent umbilical cord mesenchymal stem cells.
  • DP20CQ formulations outperformed DP80CH formulations in transfection efficiency across four donor-derived cell types.
  • The highest transgene expression with DP20CQ formulations exceeded that of the commercial reagent Lipofectamine.
  • DP20CQ formulations demonstrated favorable physicochemical properties for effective gene delivery and promoted endosomal escape.
