MicroRNAs (miRNAs) are critical regulators of osteoarthritis (OA) pathogenesis, with miR-140-5p showing the most extensive validation in cartilage protection.

  • Non-coding RNAs (ncRNAs) modulate gene expression at various levels and influence OA progression through multiple biological processes.
  • Key signaling pathways affected by ncRNAs in OA include NF-κB, TGF-β/SMAD, Wnt/β-catenin, PI3K/Akt, and MAPK/ERK.
  • miR-140-5p suppresses extracellular matrix degradation mediated by ADAMTS5 in various experimental systems.
  • Other miRNAs like miR-146a, miR-21, and miR-34a are involved in regulating inflammation and chondrocyte survival through specific pathways.
  • Many lncRNAs and circRNAs lack extensive validation in human OA tissues despite being studied in cell-based models.
