New 2-substituted benzooxazole compounds targeting multiple pathways for Alzheimer's disease treatment

Abstract

The optimal compound 92 showed a potent AChE inhibition activity with an IC of 0.052 ± 0.010 μM.

  • Compound 92 also inhibited BuChE activity with an IC of 1.085 ± 0.035 μM.
  • It significantly reduced amyloid-beta aggregation at a concentration of 20 μM.
  • The compound demonstrated good blood-brain barrier permeability at 10.80 ± 0.055 × 10 cm/s.
  • In SH-SY5Y neuroblastoma cell lines, compound 92 exhibited neuroprotective properties at 40 μM.
  • Behavioral studies indicated that a 10 mg/kg dose of compound 92 improved cognitive and spatial memory in various rat models.
