A new tiny particle system that improves poultry vaccines

Abstract

Lipid nanoparticle formulas containing plasmid DNA ranged in size from 45.4 to 376.3 nm.

  • All lipid nanoparticle formulas exhibited a spherical shape under electron microscopy.
  • Smaller nanoparticle sizes were associated with lower plasmid DNA encapsulation and higher polydispersity index (PDI).
  • Formulas with larger sizes and lower PDI values were able to encapsulate higher concentrations of plasmid DNA.
  • The high-payload vaccine formula maintained stability at 25 °C and 4 °C for up to 6 weeks, although gene expression decreased after this period.
  • Immunization with the QTAP nanoadjuvant system reduced viral shedding of avian Infectious Bronchitis Virus (IBV), despite low concentrations of plasmid DNA.
  • Pre-challenge sera from immunized chickens did not show IBV-specific antibodies when compared to those immunized with a commercial live-attenuated IBV vaccine.
