Targeting aging cells linked to NOX4 with iron-removing treatment to slow spine disc degeneration

Updated

Abstract

NOX4-associated ferro-aging is linked to intervertebral disc degeneration in chronic low back pain.

  • Ferro-aging in degenerative nucleus pulposus cells (NPCs) is associated with iron overload, oxidative stress, and cellular senescence.
  • Clinical imaging and transcriptomic analyses revealed consistent ferro-aging changes in human disc specimens and aged rats.
  • NOX4 is positioned as an upstream regulator of oxidative stress that affects lipid peroxidation and NPC senescence.
  • Amentoflavone was identified as a compound that interacts with NOX4 and suppresses NADPH oxidase activity.
  • SenoFAN, engineered with iron-chelating properties and NKG2D-engineered NPC membranes, demonstrated reduced oxidative stress and improved mitochondrial function in vitro.
  • In a rat model, SenoFAN reduced iron deposition, preserved disc structure, and exhibited favorable biosafety.
