Lipid kinase class 3 phosphatidylinositol 3-kinase (PI3K-3) is a co-activator of epigenetic writers that promotes RNA polymerase II transcription.

  • Nutrient levels may influence RNA polymerase II and epigenetic writers, impacting transcriptional processes.
  • PI3K-3 is associated with transcriptionally engaged RNA polymerase II that is phosphorylated at Ser5.
  • Nuclear PI3K-3 interacts with RNA polymerase II and the Setd1a/COMPASS complex, enhancing their binding to chromatin.
  • Loss of PI3K-3 may reduce levels of RNA polymerase II phosphorylated at Ser5 and the H3K4me3 mark at specific genes.
  • Overexpression of PI3K-3 could enhance co-activation of p300/CBP and increase H3K4me3 levels.
  • During starvation, PI3K-3 may induce genes related to autophagy and facilitate the liver's transition to ketogenesis and lipid degradation.
