Movement of USP15 between the cell nucleus and cytoplasm promotes harmful cell recycling that disrupts neural stem cell balance and brain development

Updated

Abstract

Three de novo variants in the USP15 gene were identified in human probands with various brain malformations and metabolic phenotypes.

  • USP15 variants altered its activity and localization within cells.
  • Dynamic localization of USP15 was observed in neural stem cells of the embryonic mouse cerebral cortex.
  • A knock-in mouse model with the p.Met978Val variant displayed abnormal cytoplasmic accumulation of USP15, impairing neural stem cell self-renewal and differentiation.
  • Impairments led to reduced neuronal output and enlarged lateral ventricles in the mouse model.
  • USP15 was found to deubiquitinate the autophagy regulator ATG16L1, affecting its turnover and autophagy processes.
  • Disruption of USP15's function may influence the balance of cellular processes, contributing to neurodevelopmental disorders.
