Abstract

OPL-7 and OPL-8 LNPs induced significantly lower anti-PEG IgM responses compared to ALC-0159 LNPs.

  • mRNA vaccines delivered via lipid nanoparticles (LNPs) have shown clinical success against COVID-19.
  • Concerns about PEG-lipid immunogenicity have led to the exploration of biodegradable alternatives.
  • Oligo(ethylene glycol)-functionalized polycarbonate lipids (OPLs) were synthesized to replace PEG-lipid.
  • OPL-7 and OPL-8 demonstrated desirable physicochemical properties and superior mRNA transfection efficiency in cell models.
  • Enhanced cellular uptake of OPL LNPs was observed, primarily through clathrin-mediated endocytosis.
  • In vivo studies indicated that OPL-7 and OPL-8 LNPs resulted in higher lymph node mRNA transfection ratios and robust immune responses without significant toxicity.
