Improved fat-based particles for delivering gene-editing tools targeting a common lung cancer mutation

Updated

Abstract

Essence

Pulmonary lipid nanoparticles may enable localized CRISPR/Cas9 disruption of KRAS G12S in lung cancer models.

Evidence

This formulation and preclinical experiment optimized Cas9 mRNA/sgRNA LNPs, then tested editing, mucus transport, toxicity, and tumor effects in A549 cell systems and mouse lung tumor models.

Caveat

Tumor growth suppression was modest and the evidence remains preclinical, so human efficacy and safety are unknown.
