Abstract

Orforglipron demonstrates neuroprotective properties in human-derived neuronal and microglial cell lines.

  • Orforglipron activated cAMP signaling, which is involved in cellular communication and function.
  • It protected neuronal cells from damage caused by excessive glutamate and oxidative stress, both common in neurodegenerative diseases.
  • Orforglipron reduced the secretion of proinflammatory proteins (IL-6, IL-8, MCP-1) in response to inflammatory stimuli.
  • In conditions of insulin resistance, orforglipron increased the activation of protein kinase B (Akt), suggesting an additional protective mechanism.
  • The brain uptake of orforglipron was low, comparable to peptide-based GLP-1 drugs, which may limit its effectiveness in treating neurological conditions.
