Abstract

OPG is essential for human endometrial stromal decidualization through its interaction with syndecan-1 to decrease Akt phosphorylation.

  • OPG was expressed in both the human endometrium and in vitro decidualized stromal cells.
  • Loss of OPG impaired the expression of IGF-binding protein-1 and prolactin, which are markers of decidualization.
  • Knockdown of OPG led to abnormal activation of Akt during decidualization.
  • Inhibition of Akt activation rescued the impaired expression of decidual markers in response to OPG loss.
  • Syndecan-1 was identified as a receptor candidate that interacts with OPG during decidualization.
  • Lower levels of OPG and Syndecan-1 were observed in decidual samples from women with recurrent pregnancy loss compared to those with normal pregnancies.
