Abstract

A subpopulation of p21+ senescent β-cells was identified early in the progression of type 2 diabetes.

  • Senescent β-cells show loss of cellular function and altered secretory profiles.
  • These cells demonstrate reduced responsiveness to glucose and increased basal insulin secretion.
  • Senescence-associated secretory phenotype (SASP) factors from p21+ β-cells can induce dysfunction in neighboring cells.
  • Janus kinase inhibitors (JAKi) may reverse secondary senescence and restore β-cell function.
  • The findings suggest a significant role for p21+ β-cells in the development of type 2 diabetes.
