Paeoniflorin reduces alcohol-related depression in mice by affecting the gut-brain connection and lowering inflammation

Abstract

Paeoniflorin (PF) significantly alleviated depressive-like behaviors in a mouse model of chronic alcohol exposure.

  • PF is associated with reduced hippocampal damage and oxidative stress in mice subjected to chronic alcohol exposure.
  • Treatment with PF restored levels of monoamine neurotransmitters typically altered by alcohol consumption.
  • PF suppressed microglial activation and inhibited the NF-κB/NLRP3 inflammasome pathway, which is linked to neuroinflammation.
  • The treatment resulted in a reshaping of gut microbiota composition, notably decreasing Rikenellaceae and Prevotellaceae families.
  • PF modulated metabolite profiles, including an increase in the neuroprotective metabolite niacin.
