Senescent Bone Cell Signals Linked to Localized Tissue Changes and Stiffness in Human Brain Models

Updated

Abstract

Chronic exposure to senescent osteocyte-derived signals is associated with localized extracellular matrix-related remodeling in human brain tissue.

  • Distinct mechanical features were observed in human cerebral organoids, including lower apparent modulus and regions of elevated modulus in cryosectioned specimens.
  • Spatially heterogeneous biomechanical remodeling coincided with upregulation of pathways related to extracellular matrix, inflammation, and cellular aging.
  • Suppression of neurodevelopmental and synaptic gene networks was noted alongside the biomechanical changes.
  • These pilot-scale findings were based on a single iPSC line and a single batch of conditioned medium, with limited experimental replicates.
