Tiny fat-based particles designed with peptides using exendin-4 as a model targeting molecule

Abstract

Exendin-4-functionalized nanoemulsions (ex-4 NE) exhibited higher uptake rates and larger AUC values compared to non-targeted PEG nanoemulsions.

  • Both types of nanoemulsions were efficiently taken up by 3T3-L1 pre-adipocytes and adipocytes.
  • Uptake rates were significantly influenced by the differentiation stage of the cells, with minimal impact from GLP-1R inhibition.
  • In adipocytes treated with ex-4, clathrin-mediated uptake pathways became more prominent, as indicated by reduced uptake with chlorpromazine.
  • Ex-4 NE produced stronger GLP-1R-dependent metabolic effects than free exendin-4, despite similar receptor colocalization with GLP-1R.
  • Early administration of ex-4 NE enhanced adipogenesis, while later addition reduced lipid accumulation.
  • 10 nM ex-4 NE showed effects similar to 1000 nM free exendin-4, suggesting enhanced receptor activation due to local ligand density.
