Changes in blood DNA methylation over time after ketamine-assisted therapy in major depression

Updated

Abstract

Depressive symptoms improved markedly following treatment, with mean Beck Depression Inventory-II scores declining from the severe to the mild range (p < .001).

  • Longitudinal DNA methylation changes were evaluated in 10 adults with major depressive disorder undergoing ketamine-assisted psychotherapy.
  • Of 802,595 CpG sites assessed, 20,770 met the initial screening criteria for changes after treatment.
  • After filtering for significance, 962 CpG loci were identified, including 517 gene-annotated loci.
  • Exploratory analysis revealed enrichment of Notch-related signaling pathways associated with the identified methylation changes.
  • These results suggest a structured peripheral methylation signature related to ketamine-assisted psychotherapy without establishing direct causality.
