Abstract

Pharmacological activation of STING with ADU-S100 significantly enhanced Mtb phagocytosis and intracellular clearance.

  • The enhancement in bactericidal activity is linked to an increase in autophagic flux.
  • Elevated levels of LC3-II protein were observed, indicating increased autophagy.
  • There was a significant increase in the colocalization of Mtb with lysosomal compartments.
  • Inhibition of autophagy with hydroxychloroquine reversed the observed effects.
  • Silencing of cGAS also confirmed the essential role of the STING-autophagy pathway.
