Blocking a specific cell cleanup process may improve venetoclax treatment in acute myeloid leukemia

Updated

Abstract

Genetic depletion of PINK1 reduced both basal and stress-induced mitophagy in acute myeloid leukemia cells.

  • PINK1 is identified as a central modulator of mitophagy in leukemic cells.
  • Loss of PINK1 leads to accumulation and functional impairment of mitochondria.
  • Stabilization of PINK1 during mitochondrial stress is regulated by Prohibitin 2 (PHB2).
  • Depleting or inhibiting PHB2 produces effects similar to PINK1 loss.
  • Both PINK1 and PHB2 depletion significantly increase AML cell sensitivity to venetoclax.
  • Targeting the PHB2/PINK1 signaling pathway may enhance the effectiveness of venetoclax-based therapies in AML.
