PHD3 helps activate HIF-1α in spinal disc cells independently of the PKM2-JMJD5 pathway

Updated

Abstract

PHD3 mice (12.5 mo old) showed increased incidence of intervertebral disc degeneration.

  • PHD3 suppression led to decreased hypoxic activation of the C-terminal transactivation domain of HIF-1α.
  • Expression levels of HIF-1α targets such as VEGF-A, glucose transporter-1, and lactate dehydrogenase A were reduced in PHD3 mice.
  • Manipulation of PKM2 and JMJD5 levels did not affect HIF-1 activity in nucleus pulposus cells.
  • There was no association found between HIF-1α and PKM2 in nucleus pulposus cells.
