Common Cell Culture Ingredients Affect Human Stem Cell Behavior and Development Differently in Male and Female Cells

Abstract

Phenol red and fetal bovine serum significantly impact the health and differentiation potential of human mesenchymal stromal cells (hMSCs).

  • Estrogens are global regulators of cellular signaling pathways that influence tissue remodeling and homeostasis.
  • The presence of estrogen-mimetic compounds in traditional cell culture media could bias studies, especially regarding sex as a biological variable.
  • Both phenol red and fetal bovine serum were found to affect proliferation, metabolism, senescence, and differentiation capacity of male and female hMSCs.
  • Dosing with estrogen at levels found in fetal bovine serum did not recover most health metrics of hMSCs.
  • Senescence and mRNA transcripts for RUNX2 and PPARG were the only outcomes not significantly different based on donor sex.
