Role of PINK1-Parkin Controlled Removal of Damaged Mitochondria in Septic Acute Kidney Injury

Abstract

LPS stimulation significantly increased LC3-II protein expression to 1.72 ± 0.05 at 4 hours in human renal tubular epithelial cells.

  • Mitophagy-related proteins LC3-II and BECN-1 were significantly elevated in response to bacterial lipopolysaccharide (LPS) in both in vitro and in vivo models.
  • In vivo, LC3-II and BECN-1 protein levels peaked at 2 hours after septic injury, indicating a robust mitophagy response.
  • Mitochondrial deformation and clearance via mitolysosomes were observed in renal tubular epithelial cells from septic rats.
  • PINK1 knockdown decreased LC3-II expression, while PINK1 overexpression increased it, suggesting a regulatory role in mitophagy.
  • PINK1 overexpression reduced proapoptotic protein levels, whereas depletion increased these levels, indicating a link between mitophagy and cell survival in septic conditions.
