Phosphatidylserine on tolerance-promoting nanoparticles is linked to oral immune tolerance

Updated

Abstract

Phosphatidylserine (PS) structure significantly influences immune responses, impacting the effectiveness of oral tolerance.

  • Different PS variants with altered structures were formulated in lipid nanoparticles to assess their tolerogenic potential.
  • LysoPS successfully induced oral tolerance, while double-chain PS did not, indicating structural differences affect immune responses.
  • Modifications in PS structure altered its surface exposure on nanoparticles, influencing immune responses to the model antigen ovalbumin (OVA).
  • Increased PS surface exposure was associated with lower OVA-specific IgG1 antibody levels and higher frequencies of T helper 3 cells (Th3).
  • These findings suggest a structure-function relationship in PS-mediated oral tolerance, guiding the design of PS-based immunotherapies.
