Physical, chemical, and biological testing of a pharmaceutical-grade lipid for reliable large-scale production of RNA-loaded lipid nanoparticles

Updated

Abstract

YCE-C10 LNPs exhibited sub-100-nm particle sizes with high encapsulation efficiencies and consistent batch-to-batch reproducibility.

  • Critical quality attributes evaluated included particle size, encapsulation efficiency, and mRNA integrity.
  • YCE-C10 LNPs showed high in vitro transfection activity comparable to a benchmark formulation.
  • Reproducibility across four independent GMP manufacturing runs had relative standard deviations below 5% for all quality attributes.
  • LNP encapsulation maintained mRNA integrity across temperatures of 25-85°C and improved cooling reversibility compared to naked mRNA.
  • A quantitative method confirmed consistent lipid composition in the formulated LNPs.
