Blocking PLK1 slows growth and increases radiation sensitivity in medulloblastoma cells

Abstract

PLK1 mRNA is overexpressed in some, but not all, medulloblastoma patient samples compared to normal cerebellum.

  • Inhibition of PLK1 by RNA interference significantly decreased medulloblastoma cell proliferation and increased cell apoptosis.
  • The small molecule inhibitor BI 2536 inhibited cell growth and colony-forming ability of medulloblastoma cells at low nanomolar concentrations.
  • BI 2536 pretreatment sensitized medulloblastoma cells to ionizing radiation.
  • Inhibition of PLK1 impaired tumor sphere formation in medulloblastoma cells.
  • Decreased expression of SOX2 mRNA in tumor spheres was observed following PLK1 inhibition, suggesting a role in targeting tumor-initiating cells.
