Adding a PolyA tail improves DNA stability and helps lab-made mRNA produce proteins better

Updated

Abstract

Essence

Segmented polyA tails stabilized template DNA and, for A30(CA15)11, raised protein production up to six-fold versus A90-tailed mRNA.

Evidence

Formulation/platform experiments tested 15 modified polyA variants in plasmid amplification, mRNA cell-culture assays, comparator tails, and selected in vivo validations.

Caveat

Only selected sequences were validated in vivo, and clinical durability, safety, and therapeutic performance were not tested.
