Abstract

The size-exclusion chromatography method enables high-resolution analysis of lipid nanoparticle (LNP) size distribution in its native solution condition.

  • LNPs evaluated ranged in size from 60-140 nm, showing significant batch heterogeneity.
  • The method achieved efficient and highly reproducible separation of LNPs.
  • Comparison with asymmetric field flow fractionation revealed similar size distribution results.
  • Two LNPs with similar bulk properties displayed profound differences in size and molecular weight distribution.
  • One LNP exhibited a narrow distribution while the other was polydisperse, indicating complexity in LNP drug products.
  • The developed method could assist in ensuring quality and consistency in LNP pharmaceuticals.
