Ponicidin may improve Alzheimer's disease by reducing brain inflammation and cell death caused by RIPK1

Abstract

Pon was found to bind RIPK1 with high affinity (K = 135 nM).

  • Pon may inhibit the release of pro-inflammatory cytokines IL-6 and TNF-α in microglial cells.
  • In neurons, Pon could suppress necroptosis by blocking the RIPK1-RIPK3-MLKL cascade.
  • Behavioral analysis indicated that Pon treatment significantly improved cognitive function in a mouse model of Alzheimer's disease.
  • Pon treatment was associated with a reduction in amyloid-beta plaque deposition and alleviation of neuroinflammation in the brain.
  • The findings suggest Pon may have dual neuroprotective effects by targeting RIPK1.
