Predicting and controlling the size of lipid nanoparticles without polyethylene glycol using experimental design

Abstract

Uniform lipid nanoparticles (LNPs) were formed with controlled particle size and polydispersity index (PdI) using a predictive model.

  • PEG-free LNPs containing a pH-responsive lipid derivative were successfully prepared.
  • The lipid solution ratio was identified as the most critical factor influencing particle size and PdI.
  • ζ-potential and siRNA encapsulation efficiency were not influenced by the preparation conditions.
  • A predictive model based on design of experiment (DoE) accurately forecasted the particle size of the LNPs.
