Liver growth linked to Pregnane X receptor activation in healthy people and the role of AKT-MTOR pathway in mice

Abstract

One-week rifampicin treatment caused a 2.7% mean increase in liver volume-to-body weight ratio in human volunteers (n=16).

  • Activation of the Pregnane X receptor (PXR) is associated with liver growth in male mice.
  • Significant liver enlargement was observed in male mice after just 1-day treatment with pregnenolone-16α-carbonitrile (PCN).
  • Four-day PCN treatment improved glucose tolerance in male mice.
  • RNA sequencing revealed upregulation of genes related to cell proliferation in male mice.
  • Increased activity of the AKT-mTOR pathway was detected in male mice following PXR activation.
  • No significant liver enlargement or YAP activation was observed in female mice.
