Abstract

Neurons in the prelimbic cortex that project to the nucleus accumbens core uniquely expressed Fos during cue-induced cocaine seeking, while those projecting to the nucleus accumbens shell did not.

  • Glutamate inputs to the nucleus accumbens facilitate conditioned drug-seeking behavior primarily from the medial prefrontal cortex, basolateral amygdala, and ventral subiculum.
  • Fos expression indicates neural activity and was observed in specific neuron populations during cue-induced cocaine seeking but not during sucrose seeking.
  • Activation of the prelimbic cortex to nucleus accumbens core pathway positively correlated with cocaine-seeking behavior, unlike the basolateral amygdala or ventral subiculum inputs.
  • Blocking dopamine signaling in the prelimbic cortex and glutamate signaling in the nucleus accumbens core attenuated cue-induced cocaine seeking but did not affect sucrose seeking.
