Phosphorylation of PRKN Ser131 may worsen cigarette smoke’s impact on cell cleanup and aging by activating the MKK3/p38 stress pathway

Updated

Abstract

Cigarette smoke extract exposure increased MKK3 expression and activated the p38 MAPK pathway, leading to phosphorylation of PRKN at Ser131.

  • Phosphorylation of PRKN at Ser131 was associated with reduced mitophagy activity and increased mitochondrial reactive oxygen species (ROS).
  • Knockdown of MKK3 and modulation of mitochondrial quality-control pathways improved mitophagy and reduced senescence markers in cells exposed to cigarette smoke extract.
  • Cells expressing the PRKN S131A mutant showed enhanced mitophagy flux and lower levels of ROS compared to those with wild-type PRKN.
  • Emphysematous lungs exhibited elevated MKK3 and senescence markers, along with decreased expression of PRKN and PINK1.
