How protein production changes over time from mRNA, saRNA, and pDNA delivered alone or together in the same lipid nanoparticle

Updated

Abstract

Lipid nanoparticles (LNPs) with different nucleic acid payloads produced distinct expression profiles in mice.

  • mRNA resulted in high, transient expression levels following intramuscular and intravenous administration.
  • Self-amplifying RNA (saRNA) led to delayed and sustained expression.
  • Plasmid DNA (pDNA) enabled prolonged low-level expression.
  • Combined-payload formulations exhibited expression profiles reflecting the contributions of individual payloads.
  • Minimal differences in physicochemical properties were observed across LNP formulations.
