Protein Kinases in Obesity and Treatments Targeting Them

Updated

Abstract

Obesity is associated with impaired insulin signaling largely mediated by oxidative and endoplasmic reticulum stress pathways.

  • Protein kinases and dual-specificity phosphatases regulate various cellular processes, including those involved in obesity.
  • Activation of the JNK pathway, triggered by oxidative and ER stress, suppresses insulin biosynthesis.
  • AMPK and mTOR play critical roles in glucose metabolism regulation at hormonal and cellular levels.
  • Certain pharmacological agents, like nifedipine, can stimulate lipogenesis and exacerbate insulin resistance, while metformin may alleviate these effects.
  • Obesity can increase oxidative stress and reactive oxygen species, promoting PKC-β activation and enhancing lipotoxicity.
  • Liraglutide and other compounds may protect against mitochondrial dysfunction and inflammation associated with obesity.
