Cell structure and function

Cell stress changes mitochondrial amounts and placement of an antioxidant protein during cell aging

Updated

Abstract

SOD2 protein levels in the mitochondrial fraction decreased early during stress-induced premature senescence.

  • Proteostasis decline is linked to stress-induced premature senescence (SIPS) characterized by mitochondrial dysfunction.
  • Both the proteasome inhibitor MG132 and the lysosomal inhibitor bafilomycin A1 induced cellular senescence and reduced SOD2 in mitochondria.
  • SOD2 mRNA and total cellular SOD2 protein levels did not decrease, indicating a change in localization rather than protein abundance.
  • Immunofluorescence analysis showed SOD2 dissociated from mitochondria, confirmed by quantitative colocalization analysis revealing reduced overlap with mitochondria.
  • Mislocalized SOD2 was often associated with aggregate-like structures and high-molecular-weight SOD2 complexes during proteostasis stress.
