How Psilocybin May Work at Different Levels and Its Effectiveness in Treating Depression

Updated

Abstract

Of 949 records screened, 46 original studies were included in the systematic review of psilocybin's antidepressant effects.

  • Psilocin, the active metabolite of psilocybin, may produce antidepressant effects through multiple mechanisms.
  • Activation of the 5-hydroxytryptamine receptor 2A (5-HT2a) receptor is one potential mechanism of action.
  • Increased brain-derived neurotrophic factor (BDNF) signaling and enhanced neurogenesis in the hippocampus are also associated with its effects.
  • Psilocybin has shown rapid and sustained symptom relief in treatment-resistant depression, particularly at a dose of 25 mg.
  • Clinical efficacy of psilocybin may be comparable or superior to traditional antidepressants like escitalopram.
