Abstract

Psilocybin produced a male-specific, enduring restoration of social behavior in SAPAP3 knockout mice.

  • SAPAP3 knockout mice display social abnormalities linked to disruptions in social cognition.
  • A 1 mg/kg intraperitoneal injection of psilocybin was administered to both male and female SAPAP3 knockout and wild-type mice.
  • Behavioral assessment seven days post-injection showed improved social interaction specifically in male SAPAP3 knockout mice.
  • Psilocybin increased neural activation in the medial prefrontal cortex and hippocampus across genotypes, with infralimbic activation correlating with improved social behavior in males.
  • No changes in neural activation were observed in the nucleus accumbens, basolateral amygdala, or dorsal striatum following psilocybin treatment.
