Abstract

Psychedelics may offer potential therapeutic benefits for depressive symptoms in schizophrenia spectrum disorders (SSDs).

  • Serotonergic psychedelics have shown rapid and sustained antidepressant effects in treatment-resistant depression.
  • Both schizophrenia and major depressive disorder are linked to disturbances in dopamine, glutamate, and neuroplasticity.
  • Schizophrenia is characterized by widespread neural connectivity issues and altered dopamine signaling.
  • Depressive symptoms in SSDs may reflect a combination of depressive-like connectivity and schizophrenia-related dysconnectivity.
  • Preclinical studies indicate psychedelics can enhance dendritic spine density and modulate network dynamics.
  • Current clinical evidence is limited, with only one ongoing early-phase trial for MDMA in schizophrenia.
