PTI-125 lowers Alzheimer's disease markers in patients

Abstract

PTI-125 treatment significantly reduced biomarkers of Alzheimer's disease pathology, neurodegeneration, and neuroinflammation in cerebrospinal fluid and plasma.

  • All 13 patients showed a biomarker response to PTI-125 after 28 days of treatment.
  • Total tau, neurogranin, and neurofilament light chain decreased by 20%, 32%, and 22%, respectively.
  • Phospho-tau (pT181) decreased by 34%, indicating suppression of tau hyperphosphorylation associated with Aβ42 signaling.
  • Cerebrospinal fluid biomarkers of neuroinflammation decreased by 5-14%, while Aβ42 levels increased slightly, consistent with treatment effects.
  • Target engagement was demonstrated by a shift in filamin A's conformation from 93% aberrant on Day 1 to 40% on Day 28.
