Raloxifene in solid lipid nanoparticles with improved drug delivery: development, lab testing, and studies of body absorption

Updated

Abstract

The optimized solid lipid nanoparticles (SLNs) of Raloxifene hydrochloride (RLX) exhibited a mean particle size of 109.7 nm.

  • The SLNs demonstrated a zeta potential of -13.7 mV and a polydispersity index (PDI) of 0.289.
  • In vitro drug dissolution studies indicated a Fickian release with a release exponent of 0.137.
  • Cell-based assays showed 6.40-, 5.40-, and 3.18-fold improvements in cytotoxicity, apoptosis, and cellular uptake, respectively, compared to pure RLX.
  • Pharmacokinetic studies in rats indicated a 4.06-fold improvement in peak concentration and a 4.40-fold increase in area under the curve (AUC) for RLX-SLNs compared to the pure drug.
  • A high degree of linear correlation was established between in vitro drug dissolution and in vivo drug absorption for both formulations.
