Designing Gene Control Regions to Improve Gene Activity

Updated

Abstract

The optimal length of the single-stranded spacer in the 5' UTR for mRNA stability is 25-30 nucleotides.

  • An optimal GC content of 32% in the spacer resulted in the highest levels of green fluorescent protein (GFP) production.
  • Insertion of a specific G-quadruplex structure in the 3' UTR significantly enhanced protein expression.
  • Carefully engineered 5' and 3' UTRs improved gene expression through mechanisms involving mRNA stability.
  • The presence of 5 × Corn in the 3' UTR likely promotes the formation of mRNA aggregates.
