Relaxin-2 helps protect brain connections through inflammation and cell recycling pathways in the memory area in an age-related Alzheimer’s model

Updated

Abstract

Neuroinflammation markers were significantly elevated in amyloid-beta-injected mice, with aged mice showing a greater increase than young mice.

  • RLN2 treatment significantly reduced IL-1β and TNF-α levels in young mice, indicating a potential neuroprotective effect.
  • In aged mice, RLN2 treatment had limited effectiveness, with persistent inflammatory activation and minimal improvement in synaptic recovery.
  • RLN2 inhibited NLRP3 signalling and reduced mTOR pathway activity, suggesting its role in modulating neuroinflammation and autophagy.
  • Cognitive and memory performance improved in young mice treated with RLN2, as measured by decreased escape latency in the Morris Water Maze and increased discrimination in the Novel Object Recognition test.
  • The findings indicate that RLN2 may offer therapeutic benefits for early-stage Alzheimer disease, particularly in younger individuals.
