Repeated Pressure Reduces Cell Recycling and Activity in Human Osteoarthritis Cartilage Cells

Abstract

Repetitive mechanical loading for 60 or 180 minutes significantly decreased the expression of autophagy-related proteins ATG5, Beclin-1, and Parkin in chondrocytes.

  • Physiological mechanical stress may impair the autophagy process in chondrocytes.
  • Chondrocytes under repetitive loading showed reduced numbers of autophagosomes and autolysosomes.
  • Degenerated organelles that did not undergo autophagy were observed in chondrocytes subjected to mechanical stress.
  • Decreased expression of mitophagy regulator Parkin was noted alongside reduced autophagy.
  • These findings suggest that mechanical stress may lead to the accumulation of defective mitochondria and abnormal proteins, potentially contributing to osteoarthritis progression.
