Mechanisms of ageing and development

Resveratrol helps liver cells resist aging caused by harmful natural toxins

Updated

Abstract

Exposure to a liver-derived toxic metabolite cocktail induces a distinct state of cellular senescence termed senescence induced by liver toxins (SILT) in both HepG2 cells and primary hepatocytes.

  • SILT features loss of proliferative capacity and proinflammatory changes in liver cells.
  • Primary human and porcine liver cells exhibit SILT when exposed to concentrations of endogenous toxins typical of liver disease.
  • SILT is associated with reduced albumin production and urea synthesis in liver cells.
  • Transcriptomic profiling of SILT shows activation of stress and inflammatory pathways alongside repression of metabolic functions.
  • Gene Set Enrichment Analysis indicates SILT is present in diseased liver tissue, unlike healthy liver.
  • Resveratrol treatment may reverse SILT effects in a species- and function-dependent manner.
