A combined resveratrol and hydrogen sulfide treatment targeting multiple causes of allergic asthma

Abstract

R-TBZ, a hybrid compound of resveratrol and a hydrogen sulfide donor, remained over 95% intact after 24 hours under neutral and acidic conditions.

  • R-TBZ showed superior efficacy in allergen-challenged bronchial epithelial cells compared to resveratrol or 4-hydroxythiobenzamide alone.
  • It enhanced mitochondrial antioxidant defenses, reduced mucus production, and suppressed pro-inflammatory cytokines such as IL-6 and TNF-α.
  • R-TBZ inhibited allergen-induced changes in epithelial cells and effectively reduced markers of fibroblast activation.
  • In a mouse model of allergic asthma, R-TBZ improved lung function and reduced airway hyperresponsiveness.
  • R-TBZ also attenuated eosinophilic inflammation and key cytokine production, alongside reversing airway structural remodeling.
